If a Cache Appliance is relocated to a new location and the IP address has changed, is it necessary to remove and re-add it to TPAM?

No. You simply need to update the address on the configuration page in TPAM and it should not be necessary to re-enroll the Cache.
In the event that you do experience any problems after relocation, the suggested workaround in that case would be to try removing and re-adding the Cache Server.
